Influenza Pyrosequencing

Synonym(s): Pyrosequencing, Antiviral resistance testing
Requisition Form N/A
Test Description The NA gene in pandemic influenza H1N1 positive samples is partially sequenced to detect the H275Y mutation, which is known to confer resistance to Oseltamivir.
Pre-Approval Needed If previous PCR testing has been performed and antiviral resistance is suspected on a positive H1N1 sample, please call the Infectious Disease Control Unit at 512-776-7676.
Supplemental Information Required N/A
Supplemental Form(s) N/A
Performed on Specimens from (sources) Human
Sample/Specimen Type for Testing Nasopharyngeal swabs (preferred), nasal swabs, throat swabs, nasal aspirates, nasal washes, dual nasopharyngeal/throat swabs, bronchoalveolar lavage, bronchial wash, tracheal aspirate, sputum, and lung tissue.
Minimum Volume/Size Required 0.5 mL
Storage/Preservation Prior to Shipping Keep cold or freeze.
Transport Medium Universal transport media (UTM) or viral transport media (VTM).
Specimen Labeling Tube must have two forms of identification, preferably full name and DOB.
Shipping and Specimen Handling Requirements
  • If specimens will arrive at the lab within 72 hours of collection, refrigerate at 2-8 ̊C and ship overnight on cold packs.
  • If specimens will arrive at the lab more than 72 hours after time of collection, freeze at -70 ̊C and ship overnight on dry ice.
Method Pyrosequencing
Turn-around Time N/A
Interferences/Limitations N/A
Causes for Rejection Not enough sample volume, does not meet current seasonal algorithm for testing.
Additional Information Positive pandemic H1N1 samples tested with the influenza real-time RT-PCR assay will receive pyrosequencing for investigational purposes only and results are not provided to submitters.
